Phase III trial of afatinib initiated against HER2 positive breast cancer

Drug maker Boehringer Ingelheim has announced it has initiated a pivotal global, open-label randomized phase III clinical trial to evaluate the efficacy of its targeted oral drug afatinib in patients with advanced, metastatic HER2 positive breast cancer who have undergone prior treatment with trastuzumab.

The question the trial will resolve is whether afatinib + vinorelbine extends the lives of these patients longer than trastuzumab + vinorelbine. Thus the primary endpoint in the "LUX-Breast 1" trial, as it will be called, will be progression-free survival (PFS). Secondary endpoints include overall survival (OS), safety and tolerability.

"LUX-Breast 1" joins " LUX -Lung 1", another clinical trial sponsored by Boehringer Ingelheim that is investigating the efficacy of afatinib in select patients with lung cancer.
