Brain cancer vaccine nearly doubles survival time in trials

Researchers attending the 16th Annual Meeting of The Society for Neuro-Oncology (SNO) in Orange County, CA gave a promising presentation this week on a brain cancer vaccine in clinical trials.

The trial, called ACT III, was a multi-center, single arm, Phase II clinical trial of rindopepimut (CDX-110), a product of Celldex Therapeutics, in patients with newly diagnosed EGFRvIII-positive glioblastoma (GB), an extremely aggressive brain cancer with a generally poor prognosis.

Rindopepimut targets the tumor-specific molecule epidermal growth factor receptor variant III (EGFRvIII), a mutated form of the epidermal growth factor receptor (EGFR), only expressed in cancer cells. This is a transforming oncogene that can directly contribute to the growth of a tumor. It is believed that about one third of all patients with glioblastoma express EGFRvIII.

According to the data in the presentation, rindopepimut appears to nearly double the overall survival times in these patients, from a little over a year to a median OS of just over two years.

Next up for Celldex will be ACT IV, a pivotal, randomized, blinded international Phase III study of rindopepimut.
