Novel anti-cancer agent extends survival in thyroid cancer

Preliminary data from the randomized, controlled Phase II/III FACT trial (Fosbretabulin in Anaplastic Cancer of the Thyroid) suggest that fosbretabulin (branded as Zybrestat in the US), a vascular-disruptive agent, extended overall survival in patients with anaplastic thyroid cancer compared to carboplatin and paclitaxel. However, the study involved only 80 patients, less than half of the intended 180.

In a 2009 study, fosbretabulin failed to out-perform infusion of single agent paclitaxel.

The FDA has granted fosbretabulin disodium orphan drug designation for the treatment of anaplastic thyroid cancer, medullary thyroid cancer and stage IV papillary or follicular thyroid cancer.

Fosbretabulin is believed to have two mechanisms of action: it binds to tubulin dimers in the cell which arrests mitosis, and it also disrupts a specific signalling pathway in the cell. The end result is the collapse of tumor vasculature, or the growing network of blood vessels in a tumor.

Thyroid cancer. According to SEER statistics from the National Cancer Institute, over 44,000 new cases of thyroid cancer will be diagnosed in 2010 and almost 1,700 will die from the disease.
